PROBLEM TO BE SOLVED: To efficiently immobilize nucleic acid and to obtain an improved sensitivity even if the nucleic acid to be immobilized is small by arranging a metal film, an organic substance layer, a biotin layer, and an avidin layer on a transparent layer. ;SOLUTION: A measurement chip is constituted of five layers, namely a metal film 2, an organic substance layer 3, a biotin layer 4, and an avidin layer 5 being formed on a transparent substrate 1. Generally, a material that is transparent to laser beams such as glass, polyethylenephthalate, and polycarbonate us used as the transparent substrate 1. A metal used for the metal film 2 includes gold, silver, copper, and aluminum. The organic substance layer 3 is formed by a compound with a silane-coupling agent or a mercapto group and other organic functional groups. The biotin layer 4 is formed by allowing a specific amount of biotin to contact the organic substance layer 3. By providing the biotin layer 4, a biotin labeled nucleic acid can be immobilized efficiently to the avidin layer 5.;COPYRIGHT: (C)1998,JPO
