PROBLEM TO BE SOLVED: To obtain a new purified immunoglobulin capable of being specifically reacted with a subunit antigen having a specific molecular weight of human DNA helicase 2, comprising a monoclonal antibody capable of recognizing the subunit, capable of analyzing intracellular localization of helicase in a cell cycle. ;SOLUTION: This new purified immunoglobulin comprises a monoclonal antibody which is capable of being specifically reacted with a subunit antigen having 86 kDa molecular weight of human DNA helicase 2 and of recognizing the subunit. The immunoglobulin is capable of readily visualizing and analyzing intracellular localization of a DNA helicase, which is a DNA dependent adenosine triphosphatase having activity of rewinding double strand of DNA and is indispensable in correction and restoration of DNA, in each period of cell cycle by fluorescent antibody method, etc. The immunoglobulin is obtained by immunizing a mouse against a HeLa cell in a mitotic phase, fusing a spleen cell obtained from the mouse with a myeloma cell, screening the formed hybridoma cell, selecting a cell to be strongly reacted with a cell in a mitotic phase and culture in an abdominal cavity of mouse after the cloning.;COPYRIGHT: (C)1998,JPO
