The application discloses a method and an apparatus for dispersing restriction fragment by two enzyme digestion and two direction electrophoresis, then isolating phenotype related mutant fragment by hybridization in situ of plus-minus method, then screened directly the whole gene sequence by in situ chromosome walking method with said fragment as a probe to determine modified region, promoter region and expression region motif, then screened the gene for expressing switch molecular with the probe designed with the latter, regulated the gene expression with switch molecular expressed by said gene or reagent designed from it. The invention is suitable for screening the therapeutic medicine for the most common lethal disease and promoting or increasing the expression of superior trait-related gene of economic animal and plant.
