The present invention provides a recombinant protein exhibiting RNase H enzyme activity, which is part of a polymerase (P) involved in the proliferation of human hepatitis B virus, an expression plasmid capable of producing E. coli and an RNase H enzyme protein using the present expression plasmid. In the present invention, since the RNase H enzyme protein of the present invention is stable and easy to separate and purify, including histidine labeling and the like, it can be widely used for selecting various virus growth inhibitors and RNase H enzyme inhibitors.
