The microsphere of chitosan derivatives to use a supporter to prepare a peptide or peptide labrary was prepared by a chemical treatment of chitosan. Thus, 25 g chitosan having 65 % deacetylation rate was dissolved in 1 l acetic acid, followed by addition of a solution containing 50 g sorbitan monooleate, 400 g ethanol and 70 g toluene to give a homogeneous suspended colloid solution. The suspended colloid solution was dropped in 100 g sorbitan monooleate at 40 deg.C for 48 hours to give 20 g chitosan beads in 10 l toluene and 1 l anhydrous acetic acid. The chitosan bead was stirred at 45 deg.C for 1 hour in ethanol-potassium solution to deacetylate o-acetyl group of 6-position of chitosan to give 27 g 400 mesh chitosan microsphere.
