FIELD: microbiology. SUBSTANCE: invention relates to preparing an antigen from the strain Mycobacterium paratuberculosis ВГНКИ"КА"-ДЕП. Strain is cultured on Watson-Reid medium for 28-35 days. The grown bacterial mass is washed out with distilled water, suspended in physiological solution to concentration 70-90 mg/cm3 and disrupted by ultrasonic oscillation at 22 kHz for 15-16 min. Cellular detritus is removed by centrifugation at 16 000-20 000 rev/min for 30-60 min and supernatant is preserved with thiomersal at ratio 1: 10 000. Method ensures to obtain more active and specific antigen that can be used for paratuberculosis diagnosis in ruminant animals in РИД. The use of РИД with obtained antigen ensures to detect by 1.4-9.8-fold more sick animals as compared with РСК.
