PURPOSE:To detect the presence of the infection of a mosaic disease virus within a short time by extracting and purifying protein originating from the pathogenic virus from leaves of a mosaic-infected garlic and preparing rabbit antiserum using said protein before detecting a mosaic disease infected strain according to electro-blot immunoassay (EBIA). CONSTITUTION:A leaf showing mosaic symptoms of a garlic is used as a material. Next, this sick leaf is frozen to be ground by a mortar and a pestle and 20 ml of extracting buffer is added per 5g of the fresh wt. of the leaf to sufficiently perform extraction at 4 deg.C or lower and the leaf is filtered by two gauzes to obtain a solution. Subsequently, the obtained virus purified specimen is mixed with an adjuvant to be administered to a rabbit to prepare antiserum. Next, a mosaic disease strain is detected by EBIA. That is, the crude juice of a garlic leaf is separated by electrophoresis and the protein thereof is subjected to western blotting on a nitrocellulose filter and the presence of virus infection is detected with respect to the protein transfered to the filter using rabbit antiserum.
