A process for the isolation of aerobic and anaerobic prokaryotic single cells from a mixed or pure culture is new and involves streaking a suspension of the cells onto a cover glass, drying it and drawing up single cells into the buffer-filled capillary of a micromanipulator. A process for the isolation of aerobic and anaerobic prokaryotic single cells from a mixed or pure culture comprises: (1) suspending the prokaryotes in a sterile buffer or another fluid; (2) diluting the suspension with the same buffer; (3) streaking the suspension as a thin layer onto a cover glass; (4) drying the cover glass in the air; (5) filling capillaries with buffer prior to the uptake of bacteria; (6) contacting the filled capillary with its a pointed tip with the selected prokaryote until a drop from the capillary completely engulfs the prokaryote; (7) sucking the engulfed prokaryote into the capillary; and (8) transferring the isolated prokaryote onto the surface of a solid culture medium.
