PURPOSE: To quickly, easily determine unsaturated iron binding ability in high sensitivity by adding an excess of iron to a specimen to bind the iron to transferrin, binding the free residual iron to aconitase and by measuring the resultant activated aconitase. ;CONSTITUTION: An excess of iron at a known concentration is added to a specimen such as humor to bind the iron to transferrin present in the specimen in the presence of a surfactant; thence, the free residual iron is brought into contact with apo-type aconitase inert unless iron is added to convert such aconitase to active-type aconitase. The isocitric acid produced from citric acid through being catalyzed by this aconitase is then detected using isocitric acid dehydrogenase as detective enzyme and nicotinamidoadeninedinucleotide and/or nicotinamidoadeninedinucleotide phosphate as coenzyme for the above dehydrogenase, thus accomplishing the objective quick, easy determination of the unsaturated iron binding ability for e.g. humor without the need for any skill.;COPYRIGHT: (C)1994,JPO&Japio
