PROBLEM TO BE SOLVED: To carry out isothermal culture while preventing the evaporation of water from the stopper of a BOD culture bottle having no collar and preventing the intrusion of oxygen in air into the inside of the culturing bottle in measuring BOD in an isothermal tank. ;SOLUTION: Culture bottles 1 having no collar for BOD measurement are placed in a light-shaded heat-insulated air tank 6. By spraying constant- temperature water continuously or intermittently using a water-temperature controlling water circulating apparatus 8, the temperatures of the inside of the tank and the culture bottles 1 are kept constant, and at the same time, water sealing water 13 is formed by making the upper part of the contact between the mouth of the culturing bottle 1 and the stopper wet. Thus, isothermal culture is carried out while preventing the evaporation of water from the periphery of the stopper of the culture bottle 1 and preventing the intrusion of oxygen in air into the inside of the culture bottle 1.;COPYRIGHT: (C)2000,JPO
