PROBLEM TO BE SOLVED: To provide a method for accurately determining the amount of nucleic acid sequences in a specimen without express calculation for the estimation of amplification factor by preparing plural standard specimens each containing nucleic acid control sequence and nucleic acid target sequence in known starting amounts and operating the specimen under specific conditions. SOLUTION: This method comprises step 102 wherein plural standard specimens each containing nucleic acid control sequence and nucleic acid target sequence in known starting amounts, step 104 wherein a test specimen containing nucleic acid control sequence in a known starting amount and nucleic acid target sequence in an unknown starting amount, step 106 wherein the amounts of nucleic acid control sequence and nucleic acid target sequence in the standard specimen and test specimen are amplified, step 108 wherein the indexes of the amplified amounts are determined, step 110 wherein amplification factor is evaluated from the indexes measured for the amplified amounts and step 112 wherein the starting amount of the nucleic acid target sequence in the test specimen is determined from the amplification factor and the indexes measured for the amplified amounts of nucleic acid control sequence and nucleic acid target sequence in the test specimen.
