Expandable gastroretentive therapeutic system with controlled release of active substance within gastrointestinal tract

The present invention relates to a preparation for the controlled release of active substances in the gastro-intestinal tract with a retarded passage through the pylorus, the preparation comprising a component which can expand on contact with gastric juice and is surrounded by a polymer casing which is permeable to gastric juice and active substances, whereby at least one active substance is present in the form of a multi-particle composition which releases the active substance into the gastric juice in retarded manner. A multistage production process of the above-described preparation comprises making a strip-like material for formation of a polymer casing, feeding the material in such a manner that two layers thereof are disposed side-by-side or one above another, sealing the layers of the strip-like material being so arranged in order to form separate but not fully sealed sections for filling therein a multi-particle composition, filling the sections with the multi-particle compositions, and sealing the sections by welding. Subsequently the sections are cut to obtain preparations and after rolling, folding or compressing the preparations are filled in solid gelatin capsules. The so produced preparation can be suitably used for the treatment of stomach diseases, further for reducing appetite and/or sensation of hunger.
