PURPOSE: Provided is a novel antimycotic protein isolated from the medium of Aspergillus niger which is harmless to a human body and mass cultured. The protein is excellent in antimycotic activity against variable pathogenic fungi therefore, it can be used for prevention and treatment of mycotic diseases. CONSTITUTION: A novel antimycotic protein consists of 58 amino acids represented by sequence No. 1 and its molecular weight is 6.2-7.0 kDa. The antimycotic protein shows antimycotic activity against pathogenic fungi such as Aspergillus fumigatus, Apergillus flavus, Candida albicans, Trichosporon beigelii and Saccaromyces cerevisiae and vegetable pathogenic fungi such as Fusarium oxysporum, Fusarium solani, and Aspergillus niger. The novel antimycotic protein is prepared by following steps: (a)inoculating a spore of Aspergillus niger to YPD medium containing 2% of dextrose, 1% of peptone, 0.5% of yeast extract; (b)performing shaking culture at 20 to 40C for three to five days; (c)performing Amicon ultrafiltration of supernatant of culture after removing cell line; (d)separating supernatant of culture by ion-exchange filter using CM-sepharose ion exchange column; and (e)purifying the protein.
