Isolating plasmid DNA from a microbial culture, is new. Isolating plasmid DNA from a microbial culture, is new and comprises: (1) acidifying the culture, mixing it with particles, incubating the mixture to immobilize the microbial cells on the particles, and separating the particles; (2) resuspending the cell-bearing particles, lyzing the cells, adding a neutralizing buffer, incubating the mixture, and separating and discarding the particles; and (3) adding particles to the supernatant, incubating the mixture, separating the supernatant, and eluting plasmid DNA from the particles with an elution buffer.
