PROBLEM TO BE SOLVED: To simply and rapidly obtain a DNA sample which does not contain humic acid or the like which inhibits the PCR reaction by extracting DNA derived from microorganisms existing in an environment containing humic acid by adding a water soluble humic acid-adsorbing agent upon destroying microorganisms in an aqueous solution.;SOLUTION: DNA derived from microorganisms existing in an environment containing humic acid is extracted by destroying the microorganisms in an aqueous solution containing a humic acid-absorbing water-soluble agent such as polyvinylpyrrolidone, preferably at 0.2-4.0 wt.%, adjusting a concentration of a chaotropic ion, followed by adsorbing DNA on a DNA-adsorbing agent such as fine silica powder. It is preferable to destroy the microorganisms by repeating the freezing and the melting with using a lytic enzyme such as lysozyme and an surfactant such as sodium dodecylsulfate and isooctylphenol polyethoxy alcohol.;COPYRIGHT: (C)2001,JPO
