'' 14-MEMBER MACROLYDE, COMPOUND, PROCESS TO PRODUCE A COMPOUND, 14-MEMBER MACRO-PRODUCT SYSTEM, MULTI-ENZYME PKS, NUCLEIC ACID, VECTOR, AND TRANSFORMING ORGANISM ''. A type I polyketide synthase ('' PKS '') is a complex multi-enzyme including a load domain linked to a multiplicity of extension domains. The first extension module receives an acyl initiator unit from the load domain and each extension module adds another cetid unit that can be processed (for example reduction). The applicants found that the Ksq domain possessed by some of the PKS's has decarboxylating activity, for example generating (substituted) acyl of malonyl (substituted). The CLF domain of type II PKS's has similar activity. By inserting load modules including these domains in PKS's not normally having them, it is possible to control the starter units used.
