Total mercaptans are assayed in an aqueous sample by forming a chromophoric substitution product between all the mercaptans in the sample and a reagent having general formula (I). Reduced glutathione (GSH) is assayed as a specifically-formed chromophoric thione by alkalinization from the substitution product formed in the above manner between the GSH and the reagent. The concentration of mercaptans other than GSH is determined on the basis of the difference in the concentrations obtained in the preceding assays. The assaying of mercaptans and GSH, particularly in the presence of other mercaptans, and especially in biological media, is a field of application.
