Novel and useful nucleic acids that can be used for detection ofchlorinated ethylene-decomposing bacteria and preferentially hybridize tothe 16S rRNA or rDNA of chlorinated ethylene-decomposing bacteria, and amethod of detection of chlorinated ethylene-decomposing bacteria and amethod of decomposition of chlorinated ethylene or ethane using thesenucleic acids are disclosed, wherein PCR is performed using nucleic acidcomprising 18 ~ 25 nucleotides that preferentially hybridizes to the 16SrRNA or rDNA of chlorinated ethylene-decomposing bacteria and has anyof base sequences No. 1 ~ 1 5, a base sequence that has at least 90%homology with any of these base sequences, or a base sequencecomplementary to any of these base sequences as the primer and the nucleicacid in a sample as the template and the DNA fragment that has beensynthesized is detected, and chlorinated ethylene or ethane can bedecomposed by introducing the chlorinated ethylene-decomposing bacteriadetected by this method to contaminated soil or underground water.
