PURPOSE: Provided are a method of electrochemical dense culture of a microorganism and a biosensor for analyzing organic material and BOD. The biosensor for BOD analysis can be operated without addition of the microorganism, can maintain an optimal bacterial activity depending on sewage and can determined BOD of the sewage successively by using activated bacteria contained in the sewage and sludge as a biological fuel cell. Also, the biological fuel cell is stably operated for up to 6 months. CONSTITUTION: The biosensor for BOD analysis comprises a measuring portion, a detecting portion of current, a monitoring portion recording detected current change, and optionally, a potentiostat, wherein the measuring portion is the biological fuel cell without additional mediator or transducer comprising an anode, a cathode, a conduction mediator of the anode and the cathode, and an anion switch membrane between the two pole, and wherein the cathode part comprises a sample containing electrochemically activated bacteria.
