PURPOSE: Provided are a preparation method of immobilized luminescent microorganism and a water testing method by using the microorganism to detect the pollution by heavy metals and organic solvents. The testing method is used for early detection of the pollution of the waste water flowing into the drinking water source and for checking the degree of pollution in the released water at the waste water treatment facility. CONSTITUTION: The preparation method of the immobilized luminescent microorganism consists of: recovering Photobacterium phosphoreum (KCTC2858), cultured for 16 hours at 18deg.C, from the culture medium, by centrifuging; preparing the immobilizing materials, solution of alginate and strontium chloride by adding 2.4wt.% sodium alginate to the solution containing 100wt.% of sterilized distilled water and 2wt.% of sodium chloride and by adding 8.2wt.% of strontium chloride solution to the solution containing 100wt.% of sterilized distilled water and 2wt.% of sodium chloride; making alginate cell solution by mixing the above recovered luminescent microorganism with the alginate solution and adding the above strontium chloride solution drop-wise so that cross linking structure is formed between alginate and strontium; and hardening the structure for 30 minutes. The test method comprises: putting the immobilized luminescent microorganism into the bioreactor; introducing culture medium and oxygen to the above reactor; stirring the immobilized microorganism in the above bioreactor; and mixing the continuously released culture medium containing luminescent microorganism with water test sample and measuring the intensity of luminescence to detect the pollution.
