PURPOSE: A fusion protein of tag protein and human interleukin-16, transformed E. coli capable of producing human interleukin-16 and a method for producing human interleukin-16 are provided, thereby producing the human interleukin-16 in higher yield. CONSTITUTION: The fusion protein is produced by fusing the C-terminal secretion type human interleukin-16 with C-terminal region of tag protein comprising thioredoxin region, histidine-rich region and enterokinase-digesting region. The method for producing human interleukin-6 comprises the steps of: cultivating E. coli transformed by a vector containing the fusion protein and centrifuging the cultivated medium to separate supernatant; performing HIS-bind chromatography with the supernatant to obtain an eluate; reacting the eluate with enterokinase; performing HIS-bind chromatography with the reaction product; and purifying the eluate.
