Determining nucleic acid sequence, useful for characterizing single-nucleotide polymorphisms, by incubating with probe in presence of phosphorothioate nucleotide and exonuclease
Determining a nucleic acid sequence (I), comprising incubating with a probe (II), adding at least one phosphorothioate nucleotide (III) in presence of enzyme (E1) that synthesizes a complementary sequence on the 3'-end of (II), and adding an enzyme (E2) that degrades the phosphorothioate-free complement, is new. Determining a nucleic acid sequence (I), comprising incubating with a probe (II), adding at least one phosphorothioate nucleotide (III) in presence of enzyme (E1) that synthesizes a complementary sequence on the 3'-end of (II), adding an enzyme (E2) that degrades the phosphorothioate-free complement and adding a detection molecule (IV) that interacts specifically with the complementary sequence in a way that allows detection of (I). The order of the various stages may be varied. An Independent claim is also included for a test kit for the process.
