The invented method for measuring basophile activation after stimulation with allergen for determining allergy to this allergen is characterized in that a blood sample, in which interleukin-3 has been added in an amount of 0.05 to 5?ng/ml, based on the sample volume, and a corresponding diluted allergen in an amount of 0.5 to 100 units/ml, is incubated for a period of 15 to 45 minutes at a temperature corresponding to a physiological medium, whereupon a labeled antibody anti-CD63 and a labeled antibody against a basophile surface sign are added therein at a temperature ranging from 0 to +10 degC and both in the amount of 3 to 30 microliter /100 microliter of blood, the sample is stirred and incubated for a period of 15 to 30 minutes in an ice bath. Subsequently the sample is subjected to lyophilization and then to flow cytometry. The present invention also relates to a kit for determining allergy to allergens, whereby the kit comprises per 100 tests 1 ng to 1 microgram interleukin-3, 200 ml buffer, 0.3 to 3 ml of labeled antibody anti-CD63 and 0.3 to 3 ml of labeled antibody against basophile surface sign, 4.33 microgram of N-formyl-L-methionyl-L-leucyl-L-phenyl alanine and optionally other vehicles.
