Isolation method of immunostimulating glucane from Pleurotus ostreatus, preferably from the stump by defibering, followed by bleaching by hydrogen peroxide under the temperature 15 to 25 °C during 15 to 24 hours in the environment of sodium hydroxide solution and by dehydration, consist in that the defibering preceding bleaching by hydrogen peroxide in the sodium hydroxide solution with the density of insoluble glucane 4 to 5 wt. % and dehydration, is realised in 26 hours after oyster fungus picking stored under the temperature 4 to 8 °C. Defibering is realised in environment of at least double amount of sodium or potassium carbonate with concentration 0.05 to 0.15 wt. %, in pH solution 8 to 9, during 1 to 8 min. with the formation of reaction suspension, which is bleached by hydrogen peroxide in environment sodium hydroxide solution with the concentration 0.05 to 0.09 wt. % and obtained insoluble glucane is dehydrated by ethanol, acetone, or lyophilisation.
