-45-The present invention provides a method ofidentifying a bacterial DNA sequence capable ofrecombining with DNA of a predetermined bacteriophage themethod comprises the steps of:(a) introducing a bacteriophage defensemechanism into a bacterium;(b) culturing the bacterium in the presence of afirst lytic bacteriophage to produce a bacterial culture,the first lytic bacteriophage being sensitive to thebacteriophage defense mechanism;(c) isolating from the bacterial culture a secondlytic bacteriophage which is resistant to thebacteriophage defense mechanism;(d) identifying DNA sequences present in the secondlytic bacteriophage which are not present in the firstlytic bacteriophage; and then(e) identifying a bacterial DNA sequence which ishomologous to a DNA sequence of the second lyticbacteriophage identified in step (d);wherein the identified bacterial DNA sequence iscapable of recombination with DNA of the first lyticbacteriophage.
