Preparing capture phase (A), for a target biological material (I), that comprises a modified protein (II) that binds specifically and (in)directly to (I) and that is immobilized on a support that includes reactive groups, is new. Preparation of capture phase (A), for a target biological material (I), that comprises a modified protein (II) that binds specifically and (in)directly to (I) and that is immobilized on a support that includes reactive groups. At least two peptides (III) are prepared, one comprising (II) and a sequence of at least 6 Lys at the N-terminus and a sequence of at least 6 His at the C-terminus, while the other has the opposite arrangement of oligoLys and oligoHis. Each (III) is immobilized on a support by covalent reaction between amino groups in the peptide and reactive groups on the support, then the efficiency of coupling determined and (A) chosen as the (III) which couples most efficiently to the support. Independent claims are also included for the following: (1) preparing a detection phase (B) where the support is replaced by a hapten; (2) kit for detection of (I) by ELISA (enzyme-linked immunosorbent assay), containing (A) and (B); and (3) detecting, and optionally quantifying, (I) using the kit of (2).
