The present invention is directed to identifying a gene, iroNec , from an extraintestinal isolate of E. coli, CP9. This gene was identified from CP9 by screening a library of 527 mutant derivatives of CP9 with active TnphoA fusion in human urine. Two mutant derivative CP9.45 and CP9.82 possessed increased PhoA activity in urine due to the Tnipho A insertion into iroNec . The product of this gene is an extracytoplasmic protein of 725 amino acids and can be used as vaccine against extraintestinal E. coli infections.
