A method of preparing a solution for quantification whereby a small amount of collected blood is quickly, conveniently and safely separated into plasma and a cell fraction. As a typical embodiment, citation may be made of a method of preparing a solution for quantification comprising a diluted plasma solution 5 for quantifying a test substance in the plasma with the use of collected blood 1 which involves the following steps: (1) the step of diluting the collected blood 1 with a first diluent solution 2a containing an indicator at a known concentration; and (2) the step of separating the thus diluted blood solution 3 into the plasma and a cell fraction with the use of a separation means 4 to thereby give a diluted plasma solution 5; optionally followed by (3) the step of diluting the cell fraction remaining after the preparation of the diluted plasma solution 5 with a second diluent solution 2b to thereby give a diluted cell fraction solution 6. A quantification method using a solution for quantification obtained by the method; an instrument for preparing a solution for quantification for the embodiment of the above methods; and a method of using the instrument.
