PURPOSE: Provided are a size-modifying-anchor primer, a method for RT-PCR using the size-modifying-anchor primer and a method for diagnosis using the same method, thereby diagnosing infection with a specific organism. CONSTITUTION: The size-modifying-anchor primer is a DNA fragment having an anchor region with a complementary sequence to RNA region, at 3' -end and tail region with random sequences, at 5'-end, wherein the RNA region is as a single strand during reverse transcription at 37-45 deg.C; the anchor region consists of 11-13 bases; and the tail region is not hybridized with DNA or RNA template non-specifically and forms no self-priming during PCR. The size-modifying-anchor primer has nucleotide sequence of SEQ ID NO:2(SMAC2) or SEQ ID NO:4(SMAC4).
