PURPOSE: Provided are a yeast transforming vector containing dominant nutrient-requiring gene, a yeast transformant containing it and a preparing process thereof. Therefore, the yeast transformant can be used for the improvement of conventional industrial yeasts and various microorganisms. CONSTITUTION: An adenine auxotroph S68 strain(KCTC 1018BP) is manufactured by the steps of: inducing mutation by treating a plasmid pJS8A white hydroxylamine; transforming a yeast with the plasmid pJS8A then culturing it in -UraAde medium; separating plasmid DNA from a pink or a white colony through E.coli; and treating the plasmid DNA with restriction enzymes to separate DNA and transforming a yeast strain therewith.
