METHOD FOR CONSTRUCTING CHIMERIC DNA LIBRARY USING EXONUCLEASE VII

PURPOSE: A method for constructing chimeric a DNA library using exonuclease VII is provided, thereby easily constructing the chimeric DNA library. CONSTITUTION: A method for constructing a chimeric DNA library using exonuclease VII comprises the steps of: (1) producing single stranded DNA from two or more heterogeneous double stranded DNAs; (2) digesting DNAs having the same direction as the single stranded heterogenous DNAs with DNA exonuclease VII to produce oligonucleotides; (3) repeating denaturation, hybridization and polymerization of the single stranded oligonucleotides as an inner primer and DNA polymerization inhibitor, and the heterogeneous single stranded DNA of the step (1) as a template using a terminal primer to produce a recombinant DNA strands having increased length; and (4) adding a terminal primer corresponding to both terminals of the target gene into the recombinant DNA strands for the PCR amplification thereof.
