PURPOSE: A novel functional oligosaccharide, glycosyl xylitol is prepared at high yield by using intermolecular transglycosylation of cyclodextrin glucanotransferase. Glycosyl xylitol shows proliferation effect on Bifidobacteria as increasing viability thereof and also, suppression effect on development of decayed tooth. CONSTITUTION: 1-30% (W/V) of sugar donors such as raw starch, extrusion starch, soluble starch, dextrin, maltooligosaccharide, cyclodextrin, modified starch or starch hydrolysate are used to glucosylate the same amount of xylitol. 50-10,000 unit/gram starch of Cyclodextin glucanotransferases from Bacillus marcerans, Thermoanaerobacter sp., Bacillus circulans, Bacillus stearothermophilus, Klebsiella pneumoniae, Bacillus megaterium, Bacillus ohbensis, Bacillus firmis var. alkalophilus, or alkalophilic Bacillus sp. are added to reaction mixture at 30-80°C and pH 3.0-9.0 for 24 hours. The stirring speed is 200 rpm. Xylitol glucosylated over one site is obtained at high yield. The yield of glucosylated xylitol is 27.8% for Bacillus marcerans, 28.3% for Thermoanaerobacter and 12.9% for Bacillus firmis compared with 5.1% for reported method using sucrose phosphorylase of Leuconostoc mscenteroides.
