Use of green fluorescent family proteins as affinity chromatography tags, useful for isolation of recombinant proteins, especially proteins from Anemonia sulcata
Use of proteins (I) of the green fluorescent protein (GFP) family from Anemonia sulcata, or other Cnidaria, as affinity chromatography tags, is new. (I), or its fragment, has at least 15 % identify, or at least 25 % similarity, at the amino acid level, with the GFP from Aequorea victoria, and is characterized by color or ability to fluoresce. Independent claims are also included for: (1) use of proteins (Ia) of the GFP family, or their fragments, as tags for purification of fusion proteins, in which the affinity of (Ia), either natural or introduced by mutagenesis, for immobilized substances, particularly metal ions, is exploited; and (2) use of (Ia) for immobilization of proteins and optionally their subsequent remobilization.
