Genetic modification of Amycolatopsis spp., used in producing vanillin, e.g. from ferulic acid, involves contacting culture with transforming and different DNA, magnesium and cesium chloride and polyethylene glycol
Transformation of Amycolatopsis sp. DSM 9991 (IA) or Amycolatopsis sp. DSM 9992 (IB) comprises: (1) cultivating (IA) or (IB) mycelium in a culture medium, and (2) contacting this culture with a mixture containing 0.25-10 micro g/ml transforming DNA, 0.4-0.7 M cesium chloride, 0-9 mM magnesium chloride, 30-50% (weight/volume) polyethylene glycol 1000 and 10-50 micro g/ml different DNA for 4.5-9 hours after entering the stationary phase. Independent claims are also included for: (1) transformed (IA) or (IB) produced by this process; (2) a process for producing vanillin using (IA) or (IB).
