PPROBLEM TO BE SOLVED: To provide reversed-phase liquid chromatography capable of using a mobile phase mainly comprising water regardless of the kind of a column. PSOLUTION: In the reversed-phase liquid chromatography using the mobile phase comprising 100% of water, the liquid passage of the mobile phase is stopped once and subsequently resumed to perform measurement in such a state that predetermined back pressure is applied to the outlet of the column by the back pressure device provided between the outlet of the column and a detector. Even in the measurement performed after the liquid passage of the mobile phase is resumed after once stopped, a reduction in holding time is suppressed if measurment is performed in such a state that back pressure is applied to the outlet of the column.
