PURPOSE: To obtain various kinds of laminarinases usable in the field to produce glucose and various oligosaccharides by reacting with a polysaccharide containing β-1,3-glucan. ;CONSTITUTION: The new laminarinase has the following physical and chemical properties. Substrate specificity, utilizes laminarin as a substrate: optimum pH, 5.0: pH stability, stable at pH 4-9 when maintained at 4°C) for 1 hr: optimum temperature, 40°C: temperature stability, stable at 40°C for 10min and leaving ≥90% of activity after the treatment at 50°C for 10 min: influence of metallic ions, inhibited by Hg2+, Cu2+ and Fe3+: molecular weight by SDS- PAGE method, about 27,000. The above enzyme can be produced by extracting antarctic krill and purifying the obtained crude enzyme by an affinity column treatment with immobilized laminarin and a gel-filtration treatment.;COPYRIGHT: (C)1995,JPO
