The invention provides complexes of EBNA1 and at least one EBNA1 complex polypeptide, and methods of using the same. Purified EBNA1 complexes and EBNA1 complex polypeptides are provided, as well as variants of the EBNA1 complexes such as complexes of polypeptide fragments or polypeptide fusions, and chimeric molecules comprised of polypeptide components of the EBNA1 complexes. Also provided are methods of making the EBNA1 complexes and EBNA1 complex polypeptides , methods of detecting, identifying, and purifying the EBNA1 complexes, methods of identifying compounds that modulate the EBNA1 complexes and EBNA1 complex polypeptides, and methods of treating or diagnosing EBV-mediated disorders using the EBNA1 complexes and EBNA1 complex polypeptides and compounds that modulate them.
