The present invention relates to the use of - a first nucleotide sequence encoding a first polypeptide sequence comprising a N-terminal fragment of the P1 bacteriophage Cre recombinase, said N-terminal fragment being linked to a first peptide, - and of a second nucleotide sequence encoding a second polypeptide .sequence comprising a C-terminal fragment of said Cre recombinase, said C-terminal fragment being linked to a second peptide,. said first and second peptides being able to bind together only in the presence of a specific third molecule, - or of vectors containing said first and second nucleotide sequences, - or of said first and second polypeptide sequences, for carrying out a process for regulating the activity of the P l bacteriophage Cre recombinase, in particular in the frame of processes of inactivation or activation of the expression of a selected gene, or for inducing chromosomal deletion, or reciprocal chromosomal translocation, or for exchanging one DNA cassette for another, or for obtaining transformed cells that can be used in the frame of gene therapy, or for obtaining transgenic animals.
