It is intended to provide a method of detecting a polymorphism in apolipoprotein E gene at improved accuracy and improved reliability; a method of judging the genotype of a polymorphism in apolipoprotein E gene; and a reagent. Amplified apolipoprotein E gene is cleaved by using first and second restriction enzymes respectively containing single nucleotide polymorphism sites of codons 112 and 158 in the recognition sequences thereof, and then the thus cleaved DNA is isolated and detected. A pair or primers and the restriction enzymes to be used in the gene amplification are selected so as to form a DNA fragment of about 100 bp or larger specific to each polymorphism. In the upstream primer, the 5’-end is located at a site from 60 bp or more and 236 bp or less upstream of the cleavage site of the first restriction enzyme.
