PURPOSE: A protein chip, a preparation method thereof, and a method for detecting the protein expression or protein interaction using the same are provided, thereby easily finding novel proteins using a small amount of proteins for detection. CONSTITUTION: A protein chip comprises (a) one substrate selected from glass, gold, plastic, silicone wafer and metal; (b) a calix crown derivative formed on the surface of the substrate; and (c) protein fixed on the calix crown derivative, wherein the calix crown derivative has the structure of figure 1a; and the protein is antigen, enzyme or peptide. A method for preparing the protein chip comprises the steps of: (a) treating calix crown derivative formed on the surface of a substrate selected from glass, gold, plastic, silicone wafer and metal; and (b) fixing a protein on the calix crown derivative. A method for detecting the protein expression or protein interaction comprises the steps of: (a) reacting a protein to be tested with the protein chip; (b) treating a reaction product with a fluorescent material-labeled material; and (c) detecting the fluorescence.
