PURPOSE: Recombinant cysteine proteinase isolated from Paragonimus westermani and a kit for diagnosis of Paragonimus westermani using the same are provided, thereby reducing cross-reaction with other parasites, and improving sensitivity of diagnosis. CONSTITUTION: The recombinant cysteine proteinase isolated from Paragonimus westermani is encoded by a gene having the nucleotide sequence set forth in SEQ ID NO: 1. The method for preparing the recombinant cysteine proteinase isolated from Paragonimus westermani comprises the steps of: designing primer 1 (5'-gaattcgctcccgaacgtattgac-3') which has a cysteine protease conserving region and is digested with a restriction enzyme EcoRI, and primer 2 (5'-gtcgaccatatcaccttccttatc-3') which is digested with a restriction enzyme SalI; reacting cDNA of Paragonimus westermani with the primers 1 and 2 to isolate a DNA fragment associated with cysteine protease; sequencing the isolated DNA fragment; and preparing cysteine protease using the sequenced DNA fragment. The kit for diagnosis of Paragonimus westermani comprises the recombinant cysteine protease as an antigen.
