METHOD FOR INTRAOPERATIVE LYMPH NODE ASSAY BY NUCLEIC ACID PCR AMPLIFICATION AND DETERMINING WHETHER THE PRESENCE OF ONE OR MORE OF MARKERS EXCEEDS CUT-OFF VALUE

PURPOSE: A method for intraoperative lymph node assay is provided, thereby rapidly and effectively analyzing intraoperative lymph node micrometastasis. CONSTITUTION: The method for intraoperative lymph node assay comprises the steps of: collecting a lymph node tissue sample from a patient; lysing the lymph node tissue sample containing interest RNA to produce homogenate; diluting the homogenate; contacting the homogenized sample with a substrate containing a RNA-binding material or immobilizes it; binding the sample to the substrate; removing contaminants and interferent; drying the substrate; eluting RNA from the substrate; PCR amplifying the RNA and detecting the PCR product to analyze the sample; and determining whether the presence of one or more of markers exceeds a cut-off value, wherein the method is useful for detecting micrometastasis, breast cancer metastasis or melanoma metastasis.
