PURPOSE: RNA as a template for efficient de novo RNA synthesis by hepatitis C virus(HCV) nonstructural protein 5B(NS5B) polymerase is provided. The RNA is useful for the prevention and treatment of HCV and for the production of virus replicon mass-producing HCV genome. CONSTITUTION: RNA as a template for efficient de novo RNA synthesis by hepatitis C virus(HCV) nonstructural protein 5B(NS5B) polymerase has the mutated nucleotide sequence of RNA having the nucleotide sequence of SEQ ID NO: 1, wherein the mutated nucleotide sequence is selected from the nucleotide sequences of SEQ ID NOs: 2 to 16. An HCV NS5B polymerase inhibitor comprises RNA having the nucleotide sequence selected from the group consisting of the nucleotide sequences of SEQ ID NO: 2 to 16, DNA corresponding to the RNA, or derivatives thereof, wherein the derivative is 2'-amino derivative, 2'-fluoro derivative, or 2'-O-methyl derivative.
