The invention relates to the use of lamellar vesicles based on amphiphilic products, preferably multilamellar vesicles having an internal crystal-liquid lamellar structure, as an agent intended to reduce the variability of the bioavailability of a drug administered orally or d one of its metabolites. The invention applies particularly to the field of anti-cancer chemotherapy. It also relates to new pharmaceutical compositions consisting of a dispersion or a suspension in a pharmaceutically acceptable vehicle compatible with administration by oral route of multilamellar vesicles with an internal crystal-liquid structure containing an active principle known for its activity in anti-cancer chemotherapy, in particular a product of the family of camptothecins and their derivatives or one of their metabolites, for example CPT 11 and SN 38.
