The identification of alpha-1-antitrypsin alleles with restriction enzymes has in vitro amplification of the exons III and V of the antiprotease gene, for incubation-based genotype assignation of the patient. The normal M and deficient S and Z alleles of the protease inhibitor are detected, manually or automatically.
