The invention relates to a vector for cloning an insert having blunt ends, enabling recombinant clones to be positively selected, to a method for cloning an insert in said vector, and to the use of said cloning vector especially for expressing an insert. The inventive cloning vector comprises a nucleotide sequence coding for a poison protein and a restriction enzyme site located in an essential coding region of said nucleotide sequence, said restriction enzyme generating blunt ends and being active in ligation conditions.
