PURPOSE: A method for preparing Androst-4-ene-3,17-dione(AD) and Androst-1,4-diene-3,17-dione(ADD) using a two-step fermentation process is provided, thereby rapidly carrying out oxidation and side chain degradation of sterols, and improving yield of AD/ADD using the oxidized and side chain degraded sterols. CONSTITUTION: The method for preparing Androst-4-ene-3,17-dione(AD) and Androst-1,4-diene-3,17-dione(ADD) using a two-step fermentation process comprises the steps of: inserting at least one selected from emulsified sterols and cyclodextrin-binding sterols into a medium and culturing a first microorganism which is useful in the function of oxidation and side chain degradation of sterols in the medium; and culturing a second microorganism which is useful in the function of further side chain degradation of the oxidized and side chain degraded sterols in a medium, wherein the sterol is cholesterol, sitosterol, campesterol, stigmasterol and ergosterol; the cyclodextrin is selected from alpha-cyclodextrin, beta-cyclodextrin and gamma-cyclodextrin; the first microorganism is selected from Arthrobacter sp., Nocardia sp., Fusarium sp., Rhodococcus sp., Mycobacterium sp., Microbacterium sp., Protaminobacter sp., Brevibacterium sp., Corynebacterium sp., Bacillus sp., Serratia sp., Azotobacter sp., Streptomyces sp., Alcaligenes sp., Pseudomonas sp. and mutants thereof; and the second microorganism is Mycobacterium fortuitum(ATCC 29472).
