PURPOSE: A process for improving efficiency of DNA amplification reaction is provided, thereby simplifying a procedure for deciding optimal temperature(annealing) condition and improving hybridization specificity of oligonucleotides to DNA. CONSTITUTION: The process for improving efficiency of a DNA amplification reaction comprises using a primer in which a compound is added into the 5'-terminal of the primer, wherein the compound is selected from LC-Red 705, amino group, phosphate group, biotin, DIG, DNP, TAMRA, Texas-Red, ROX, XRITC, rhodamine, LC-Red 640, mercapto group, psoralen, cholesterol, FITC, 6-FAM, TET, cy3, cy5, BODIPY 564/570, BODIPY 500/510, BODIPY 530/550, BODIPY 581/591 and oligonucleotides having a combined G and C content of at least 25% and having at least four bases.
