PURPOSE: Provided are abscisic acid responsive element-binding factors (ABFs) which bind with abscisic acid responsive elements(ABREs) to regulate transcription and are thus useful in developing transformed plants resistant to complicated environmental stress. CONSTITUTION: Abscisic acid responsive element-binding transcription factors (ABFs) are separated from Arabidopsis cDNA expression library using typical ABRE(GGACACGTGGCG) by yeast one-hybrid screening method. In particular the separating procedure is as follows: i) separating RNA from ecotype Colunbia; ii) determining bases of DNA and purifying RNA through ethanol or LiCl precipitation methods; iii) analyzing positive clones; iv) separating ABF3 and ABF4 through PCR and determining base sequences of several clones; v) making plasmid pYX243; vi) manufacturing recombinant ABF; vii) synthesizing oligonucleotide pool, R58, composed of 18 random bases to obtain DNA; viii) measuring the transcription activity of DNA; and ix) separating ABRE-binding transcription factors.
