Complete, or nearly complete, rpoB genes from Corynebacterium or related bacteria, comprising any of: (a) sequences (61)-(116), reproduced; (b) sequences with at least 98% similarity with (a); and (c) their reverse sequences and complements. Independent claims are also included for: (1) fragments of the rpoB gene from the species specified above, or from C. efficiens or C. glutamicum, that comprise any of sequences (3)-(60), reproduced, or sequences at least 98% homologous, or reverse sequences or complements; (2) oligonucleotides (ON1) containing at least 20 consecutive nucleotides (nt) from (3)-(60), their homologs, reverse sequences or complements; (3) oligonucleotides (ON2) containing at least 8, preferably 35, nt of which at least 8, preferably 18, are consecutive within the sequences (1) and (2) 5'-CGWATGAACATYGGBCAGGT (1) 5'-TCCATYTCRCCRAARCGCTG (2) where W = A or T, Y = C or T, B = C, G or T and R = A or G; (4) equimolar mixtures of different ON2 containing at least 12, preferably 18 consecutive nt from (1) or (2), or their reverse sequences or complements; (5) in vitro molecular identification of a species of Corynebacterium or related bacteria; and (6) diagnostic kit for method (5) containing at least one ON1, ON2 or the rpoB fragments.
